HRP-IGG CONJUGATION KIT

The HRP-IgG Conjugation Kit utilizes a novel chemistry to generate highly reproducible IgG-HRP conjugates with a simple procedure. The resulting conjugates have been shown to be extremely stable, retaining 100% activity after storage for 60 days at 37º C with concentrations as low as 0.5 μg/mL. Manufacturing quality systems ensure excellent sensitivity, low backgrounds and lot to-lot consistency.

This kit allows users to generate their own liquid stable conjugates that are highly stable at working concentrations, can be customized and are fully scalable from 0.01 mg to gram scale. Each kit includes all reagents needed to conjugate the stated amount of IgG at an HRP-IgG ratio of 4:1. Kits are available for conjugation of 0.2mg, 1mg , 5mg and 10mg IgG.

BACKGROUND

Enzymes have been used as detection probes for many years and are still commonly used, despite the introduction of fluorescent probes, because of their simplicity, sensitivity and broad-spectrum applications. Enzyme probes, including horseradish peroxidase (HRP) and alkaline phosphatase (AP), are used to detect target proteins through chromogenic, chemiluminescent or fluorescent outputs. HRP is widely used as an enzymatic label in immunochemistry assays. Preparing stable and reproducible antibody-HRP conjugates is one of the biggest challenges of developing immunoassays. The HRP-IgG conjugation kit utilizes a highly robust chemistry to generate highly reproducible IgG-HRP conjugates with a simple procedure. The resulting conjugates have been shown to be extremely stable, retaining 94% activity after storage for 95 days at 37º when stored at a concentration of 0.5 µg/mL. Conjugates are suitable for use on various Immunoassay such as ELISA, Flow Cytometry and Multiplexing platforms such as Luminex. For example, HRP conjugated antibodies have been used in ELISA for testing clinical serum samples for the presence of Dengue virus NS1 (Lai et al., 2019).
